In Vitro Propagation of Ficus Benjamina by Shoot Tip Culture



Abstract
The shoots of Ficus benjamina were cultured on the basal medium containing MS macroelements, Nitsch & Nitsch microelements and MS vitamins. Cytokinin, auxin, sucrose, adenine¡¤SO©þ and NaH©üPO©þ¡¤H©üO were supplemented on the medium to micropropagate Ficus benjamina. Shoot multiplication was favorable on the media supplemented with 0.5-2.0 §·/L BA or 0.01-0.05 §·/L TDZ. The best shoot multiplication was obtained on the medium with 1.0 §·/L BA and 0.1 §·/L IAA, and shoot multiplication was more enhanced on the media containing BA and IAA in combination than those with BA alone. But the formation of callus was stimulated as the concentration of IAA increased. TDZ was not effective on shoot multiplication and shoots were rooted when TDZ was supplemented with IAA. The formation of callus was inhibited and shoots were multiplicated favorably on the media with 10-20 g/L sucrose, although callus formation was enhanced and shoot multiplication was decreased on the media containing more than 25 g/L sucrose. Shoot multiplication was inhibited by the addition of BA and adenine¡¤SO©þ, but it was increased by the addition of 0.01 §·/L TDZ and 40-80 §·/L adenine¡¤SO©þ. The combined additions of NaH©üPO©þ¡¤H©üO and BA or TDZ inhibited shoot multiplication, and increased callus formation.
